Red Blood Cell Lysis Buffer is a reagent used to selectively lyse non-nucleated erythrocytes It is designed to remove erythrocyte contamination from biological samples thereby enabling the purification of nucleated cells such as lymphocytes Red Blood Cell Lysis Buffer exerts its biological activity primarily through ammonium chloride-based osmotic shock which disrupts erythrocyte membranes while sparing nucleated cells Following red blood cell lysis the resulting preparations typically contain greater than 90% nucleated cells Based on these properties Red Blood Cell Lysis Buffer is utilized in applications such as nucleic acid extraction protein isolation cell culture assays and flow cytometry analysis of samples from human mouse rat and other mammalian sources
