Perform Antimicrobial disc diffusion susceptibility testing of Neisseria gonorrhoea with Thermo Scientific™ Remel™ GC Agar Base with 1% GCHI.

In 1945, Johnston developed modified chocolate agar that allowed the growth of N. gonorrhoea in 24 hours rather than 48 hours1. Fast growth rate was traced to differences in gel strength of the media tested. Therefore, GC Agar Base w1% GCHI was developed with reduced agar content. In 1977, Thornsberry et al. used GC Agar Base to determine β-lactamase production by N. gonorrhoeae2. Jones et al. recommended adding a modified GCHI Enrichment into GC Agar Base3.

GC Agar Base w1% GCHI is a solid medium used for antimicrobial susceptibility testing of Neisseria gonorrhoea using the disc diffusion technique. The effect of penicillin is inhibited when pathogenic Neisseria produce β-lactamase. Gonococci with 10U penicillin disc zone diameters ≤ 19 mm are likely to be β-lactamase producing strains4.

GC Agar Base w1% GCHI contains casein and meat peptones that supply corn starch to neutralize toxic fatty acids. The phosphate buffer controls pH changes that can affect organism survival. The GCHI Enrichment provides NAD, vitamins, amino acids, coenzymes, dextrose, ferric ions and other growth factors needed to cultivate Neisseria species.
