Ellis et al. modified Kauffmann and Petersen's formula to prepare this medium1,2. Mucate Medium (Mucic Acid) is recommended for use in conjunction with other tests to differentiate and identify certain members of Enterobacteriaceae, including E. coli and Shigella3,4.
Mucate Medium (Mucic Acid) is a liquid medium that is used to differentiate Shigella and nonmotile Escherechia coli based on mucic acid utilization.
Mucic acid gets converted to mucate when pH of the medium is adjusted by sodium hydroxide during manufacturing process. Utilization of mucate results in an acid reaction that causes the indicator (brom thymol blue) to change color from blue to yellow.
