Presumptively identify Escherichia coli serogroup O157:H7 grown on culture media using RIM E. coli O157:H7.

Identify Escherichia coli serogroup O157:H7 grown on culture media using Thermo Scientific™ Remel™ RIM E. coli O157:H7 Latex Test. E. coli O157:H7 is difficult to detect with routine methods since most strains ferment lactose rapidly and are indistinguishable from non-O157:H7 E. coli on conventional enteric media. The selective, differential medium Sorbitol MacConkey Agar (SMAC) helps detection of E. coli O157:H7 due to their inability to ferment sorbitol. E. coli O157:H7 Latex Test can be used to identify E. coli O157:H7 from sorbitol-negative colonies grown on SMAC primary culture plates.

Escherichia coli O157:H7 was first recognized as a cause of hemorrhagic colitis in 19821,2. E. coli O157:H7 produces the verocytotoxin that has been identified as an agent of hemolytic uremic syndrome (HUS)3-5. Unlike most strains of non-O157:H7 E. coli, Salmonella, and Citrobacter, E. coli O157:H7 does not ferment sorbitol. Sorbitol-fermenters form pink to red colonies on SMAC whereas sorbitol non-fermenters are colorless.

E. coli O157:H7 Latex Test provides 3 latex reagents. The particles in each reagent are coated with a different antibody: One against E. coli serotype O157, another against E. coli serotype H7, and the third with normal rabbit globulin, to serve as the control latex.

When test latex particles are mixed with fresh colonies of O157 and/or H7 strains of E. coli, an immunochemical reaction occurs resulting in agglutination. Absence of agglutination indicates the test isolate is not E. coli O157:H7. The Control Latex reagent identifies non-specific agglutination.
