Presumptively identify Candida albicans and Cryptococcus neoformans by germ tube formation, chlamydospore production and phenol oxidase activity using Thermo Scientific™ Remel™ TOC Agar (Polysorbate 80, Oxgall, Caffeic Acid). Two media were developed for the sequential development of germ tubes and chlamydospores by C. albicans within 24-48 hours1,2 , containing a combination of rice infusion, oxygall, polysorbate 80, 0.1% glucose and 2% New Zealand Agar. Later a culture medium was developed to aid in the identification of both C. neoformans and Candida spp.3 which became known as TOC Agar.

TOC Agar is a differential medium used for the presumptive identification and differentiation of Candida albicans and Cryptococcus neoformans. It contains polysorbate 80 and oxgall which increase the rate at which germ tubes and chlamydospores are formed. Caffeic acid serves as a substrate for detection of phenol oxidase produced by C. neoformans. This enzymatic reaction produces melanin which is absorbed by the yeast cell wall resulting in brown-pigmented colonies.

In chlamydospore production clusters of round blastoconidia at the septa and large, thick walled terminal chlamydosopres are seen and in pigment production growth tan to brown-pigmented colonies are observed
