Selectively recover Salmonella species other than Salmonella enterica serovar Typhi with Thermo Scientific™ Remel™ XLT4 Agar Monoplate. XLT4 (Xylose-Lysine-Tergitol 4) Agar was developed by Miller and Tate to selectively screen samples contaminated with enteric organisms for the presence of Salmonella1. They found that XLT4 Agar completely inhibits competing enteric organisms. Mallinson et al. reported XLT4 Agar demonstrated significant improvements over traditionally used differential media for the detection of Salmonella from raw red and white meat samples and poultry litter2.

Salmonella is a rod-shaped, gram-negative facultative anaerobe that belongs to the family Enterobacteriaceae and is one of the most commonly isolated foodborne pathogens found predominantly in poultry, eggs and dairy products. Numerous Salmonella serotypes have been identified, majority of which belong to Salmonella enterica subsp. enterica, and can cause serious, at times life-threatening, infections in humans.

XLT4 Agar Monoplate is a solid medium that is used for selective and differential recovery of non-typhi Salmonella species from clinical specimens.

    • H2S-positive organisms like salmonellae produce black-yellow colonies.
    • Lysine-negative organisms e.g., Escherichia coli, Citrobacter, and Proteus spp. produce yellow colonies.

XLT4 Agar Monoplate contains meat peptone and yeast extract that supply nitrogenous compounds, amino acids and vitamins necessary for bacterial growth. Xylose is the carbohydrate source. Lysine helps to isolate salmonellae based on their ability to decarboxylate lysine and produce an alkaline pH. Lactose and sucrose are added in excess to prevent colonies of lysine-positive coliforms from reverting to alkaline conditions. Sodium thiosulfate is a sulfur source and ferric ammonium citrate is an indicator of H2S-production.

Tergitol 4 acts as a selective agent and enhances the specificity and sensitivity of Salmonella detection and reduces the growth of non-salmonellae enteric gram-negative bacilli.
