Inhibit Rho-associated, coiled-coil containing protein kinase (ROCK), including both ROCK1 and ROCK2 (Davies et al. Biochem J, 2000; Ishizaki et al. Mol Pharmacol, 2000), with Y-27632. Y-27632 enhances the survival and increases the cloning efficiency of human embryonic stem (ES) cells (Watanabe et al. Nat Biotechnol, 2007), improves embryoid body formation using forced-aggregation protocols (Ungrin et al. PLoS One, 2008), and increases the survival of cryopreserved single human ES cells after thawing (Li et al. Hum Reprod, 2009). In combination with other factors, it allows for the direct lineage reprogramming of fibroblasts to mature neurons (Hu et al. Cell Stem Cell, 2015). Y-27632 also improves survival of human ES cell monolayers at the initiation of differentiation protocols (Rezania et al. Nat Biotechnol, 2014).
