RMC-6291 is an orally bioavailable, covalent inhibitor of the active, guanosine triphosphate (GTP)-bound form of the oncogenic KRAS substitution mutation G12C, KRASG12C(ON). RMC-6291 forms a tri-complex with the intracellular chaperone protein and immunophilin cyclophilin A (CypA) and KRASG12C(ON). This tri-complex blocks KRASG12C(ON)-mediated signaling. RMC-6291 elicits deep and durable suppression on RAS pathway activity in KRASG12C tumor models (refs 1, 2).
