RP-64477 is a potent inhibitor of the cholesterol esterifying enzyme Acyl-coenzyme A:cholesterol O-acyltransferase (ACAT).

  • Potent ACAT inhibitor with IC50 values ranging from 6 to 283 nM in various animal tissue preparations, and 113 to 503 nM in human cell lines (HepG2, CaCo-2, and THP-1).
  • No inhibitory activity was observed against rat PCEH or LCAT at concentrations up to 200 μM and 20 μM, respectively.
  • Administration of RP-64477 significantly reduced plasma cholesterol levels in cholesterol/cholic acid-fed rats.
