Saline 0.45% is a sterile sodium chloride solution commonly referred to as half normal saline. With a salt concentration lower than physiological saline, it is considered hypotonic relative to most biological cells. This property allows water to move into cells through osmosis, causing them to swell under controlled laboratory conditions. Such behavior is useful in certain cytological and cell preparation procedures where swelling assists with cell lysis or chromosome spreading for microscopic examination. The solution is also used for rehydrating freeze dried reagents, enzymes, or antibiotics when lower ionic strength is required. In laboratory workflows it may also serve as a gentle washing or dilution solution in molecular biology and biochemical procedures.
