Simmons Citrate Agar is a differential culture medium used to determine whether microorganisms can utilize citrate as their sole carbon source. The medium contains sodium citrate as the only carbon source and ammonium phosphate as the nitrogen source. Bacteria capable of metabolizing citrate convert these compounds into alkaline byproducts that increase the pH of the medium. Bromothymol blue is included as a pH indicator that changes color from green to blue when alkalinity develops. This reaction indicates a positive citrate utilization result. Simmons Citrate Agar is widely used as part of the IMViC test series for differentiating members of the Enterobacteriaceae family.
