SOX2, also known as SRY (sex-determining region Y)-box 2, is a member of the Sox family of transcription factors with a highly conserved DNA-binding domain known as the HMG (high mobility group) box domain. SOX2 has been shown to play critical roles at many stages of mammalian development and regulates pluripotency in embryonic stem cells. Knockdown or dysregulation of SOX2 leads to terminal differentiation or embryonic lethality, suggesting that Sox2 is essential for embryonic development. SOX2 has been shown to be required for the development and growth of various types of tumors, but it has also been reported that SOX2 can inhibit tumor growth. These different functions of SOX2 in tumorigenesis still need to be further demonstrated by more experiments, which may help to understand the context-dependent role of SOX2 as an oncogene and tumor suppressor gene.
