SR9011 hydrochloride is a potent REV-ERBα/β agonist (IC50s: 790 nM for REV-ERBα, 560 nM for REV-ERBβ). It significantly increases REV-ERB-dependent repressor activity and suppresses transcription via full-length REV-ERBα. The compound inhibits BMAL1 mRNA expression in HepG2 cells and suppresses breast cancer cell proliferation by halting the cell cycle before M phase. In mice, SR9011 suppresses REV-ERB responsive genes and temporarily reduces locomotor activity during the dark phase.
