Tetracaine hydrochloride (Amethocaine hydrochloride) is a calcium channel protein inhibitor that blocks voltage-sensitive release of Ca2+ from sarcoplasmic reticulum. It is primarily used topically in ophthalmology and as an antipruritic. It alters the function of calcium release channels (ryanodine receptors) that control calcium release from intracellular stores, acting as an allosteric blocker of channel function.
- Inhibits spontaneous calcium release events at low concentrations
- Completely blocks calcium release at high concentrations
- Soluble in H2O (50 mg/mL, requires ultrasonic)
- Soluble in DMSO (≥ 30 mg/mL, use newly opened DMSO)
- Unstable in solutions, fresh preparation is recommended
