Thermo Scientific MagJET NGS Cleanup and Size Selection kit is designed for efficient and robust DNA fragment library cleanup and size-selection from a variety of enzymatic reaction mixtures including PCR, ligation, adapter addition and DNA end-repair reaction mixes.

The kit utilizes high-capacity MagJET paramagnetic bead technology that ensures efficient recovery of DNA fragment library within a desired fragment length range. The purified DNA fragments are free of any next-generation sequencing workflow inhibiting components, such as sequencing adapters, primer dimers, unincorporated nucleotides, enzymes or salts, and are ready-to-use in downstream NGS applications. No prior DNA fragment library purification is required before the cleanup and size-selection procedures.

The kit is suitable for cleanup or size selection of 5 ng to 5 μg of DNA fragment library. MagJET cleanup and adapter removal protocols can easily be automated for higher throughput on automatic platforms such as Thermo Scientific KingFisher Flex and Thermo Scientific KingFisher Duo.

Highlights
Robust—reliable cleanup and size selection from different reaction mixtures and buffers
Flexible—user-defined size-selection of exact DNA fragment range
NGS-grade purity—complete removal of NGS adapters, primers, nucleotides, enzymes and other reaction components inhibiting NGS workflow
High throughput compatible—easy to automate on the Thermo Scientific KingFisher magnetic particle processors for high-throughput sample handling

Applications
DNA fragment library cleanup and size-selection for downstream use in next generation sequencing workflows.

Can I use water instead of Elution Buffer in the elution step of GeneJET and MagJET nucleic acid purification kits?

We would highly recommend using the provided Elution Buffer in the elution step for best results. You may use nuclease-free water; however, it may affect the yield of the nucleic acids during purification and their stability during storage.

Are GeneJET spin columns or MagJET magnetic beads available separately?

No, we do not offer GeneJET columns or MagJET magnetic beads separately at this time.

Can I edit the manufactured KingFisher Flex protocols?

No, manufactured protocols are locked for editing, but they can be renamed using the ‘save as' function and edited as a new version.

Can I use magnetic beads from other vendors with the KingFisher Flex instrument?

For nucleic acid extraction, we recommend using our MagMAX kits and MagJET kits. Other magnetic beads should work well as long as they are 0.5 to 10 µm in size. However, you will have to write your own KingFisher protocols using the Thermo Scientific BindIt software and validate/optimize the protocol on your own.

I am working with liquid samples with high volume and want to extract nucleic acid from them using the KingFisher Flex instrument. Can I use more volume than the protocol advises?

Yes, this can be done by sample splitting. Set the protocol so that the magnet will collect samples from multiple wells before continuing to the next step. Be sure not to overload.

Can I scale up my reaction volume with the KingFisher Flex instrument?

Yes, reaction volumes can be scaled up, and all other volumes (i.e. wash volumes) will have to be increased by the same fold difference. It is generally not recommended to scale up reaction volumes more than 2-fold unless the plate's volume capacity will allow it.
