TrueBlack Lipofuscin Quencher is a novel reagent for quenching lipofuscin autofluorescence in immunofluorescence (IF) and IHC. Highly autofluorescent lipofuscin granules, products of protein and lipid oxidation, build-up in lysosomes due to tissue aging and fluoresce brightly in all channels. This makes imaging of specific IF signal in certain tissues challenging unless methods are employed to quench or mask lipofuscin fluorescence. TrueBlack effectively quenches lipofiscin autofluorescence in a variety of tissues. It also reduces autofluorescence from other sources like collagen, elastin, or RBCs. Treatment is simple, rapid, can be performed pre- or post-IF staining. It has minimal effect on fluorescent antibody or nuclear counterstain signal, nor does it increase red/far-red background unlike Sudan Black B. TrueBlack has also been used in imaging cytometry to quench autofluorescence from polycarbonate filters.
