Tryptic Soy Agar with 5% Sheep Blood is a differential culture medium used for the isolation and identification of bacteria based on their hemolytic activity. The addition of sheep blood allows visualization of hemolysis patterns around bacterial colonies. Alpha hemolysis produces a greenish discoloration, beta hemolysis results in clear zones of complete red blood cell lysis, and gamma hemolysis indicates no hemolysis. These patterns are essential for identifying clinically significant organisms such as Streptococcus species. The 14mL plate format provides sufficient surface area for colony development and observation. This medium is widely used in clinical microbiology and diagnostic laboratories.
