The TUNEL Apoptosis Detection Kit (DAB) is designed to detect nuclear DNA fragmentation occurring during apoptosis in tissues or cells During apoptosis endonucleases within cells become activated and cleave genomic DNA between nucleosomes resulting in DNA fragments of approximately 180-200 bp or multiples thereof The fragmented DNA generates free 3 -OH termini at which terminal deoxynucleotidyl transferase (TdT) incorporates biotin-labeled dUTP Subsequently these labeled termini bind to horseradish peroxidase (HRP)-conjugated streptavidin (Streptavidin-HRP) followed by chromogenic visualization using DAB This kit allows direct observation using an ordinary optical microscope and is convenient for use It is applicable to a wide range of sample types including frozen sections paraffin-embedded sections adherent cell cultures and suspended cells making it versatile for apoptosis detection
