Tunicamycin is a nucleoside antibiotic that inhibits the biosynthesis of N-linked glycoproteins. It inhibits the formation of N-glycosidic linkages during the early steps of protein glycosylation. Also, it interferes with the transfer of N-acetylglucosamine-1-phosphate to dolichol phosphate, a crucial step in the synthesis of lipid-linked oligosaccharides. As a result, the glycosylation process in the endoplasmic reticulum is disrupted.

This antibiotic induces endoplasmic reticulum (ER) stress by disrupting protein folding and glycosylation in the ER.
