The PathHunter U2OS AVPR2(S167T) ENDO-EA Pharmacotrafficking Assay is a stable, engineered cell line for use in studying pharmacochaperones targeting AVPR2. This cell line enables assessment of chaperone-based stabilization of AVPR2(S167T) via detection of rescued membrane-endosome trafficking. The included cell line overexpresses ProLink-tagged mutant AVPR2(S167T), which is misfolded and retained in the ER. The cell line also expresses endosome-localized EA. When a pharmacochaperone stabilizes the mutant AVPR2, it is trafficked to the membrane and then internalized to the endosome, producing EFC signal.
