The UbiTest kit identifies whether a target protein is polyubiquitinated and improves on traditional immunoprecipitation and Western blot methods, which often fail due to epitope masking, reduced antibody affinity, or changes in selectivity. By combining immunoprecipitation with broad‑spectrum DUB digestion, the assay reveals increased signal for the protein of interest after DUB treatment, confirming ubiquitination even when untreated samples show weak detection. To avoid variability caused by antibody–substrate interactions, UbiTest uses high‑affinity TUBEs to pull down polyubiquitinated proteins. These engineered ubiquitin‑binding domains have nanomolar affinity and TUBE1 binds all eight linkage types. The kit contains only the buffers needed for efficient elution of polyubiquitinated proteins.
