Tandem Ubiquitin Binding Entities (TUBEs) are powerful tools for isolating and characterizing polyubiquitinated proteins. They bind K48‑ and K63‑linked tetra‑ubiquitin with single‑digit nanomolar affinity—100–1000 times stronger than monomeric UBAs—and protect ubiquitinated proteins from DUBs and proteasomal degradation. This allows efficient recovery of native polyubiquitin chains from cells and tissues under gentle conditions. TUBE 1 and TUBE 2 enrich all ubiquitin linkage types, making them useful when linkage specificity is unknown. K48 TUBE HF offers high selectivity for K48‑linked chains and can be paired with K63 or M1 TUBEs to analyze chain architecture. Together, these reagents provide a robust platform for polyubiquitin isolation and linkage characterization.
