Tandem Ubiquitin Binding Entities (TUBEs) are powerful tools for ubiquitin research due to their extremely high affinity for K48‑ and K63‑linked polyubiquitin chains—100 to 1000 times stronger than monomeric UBAs. They also protect ubiquitinated proteins from deubiquitinases and the proteasome, enabling efficient isolation of native polyubiquitin chains under mild conditions that preserve cell physiology. TUBE 1 and TUBE 2 enrich all ubiquitin linkage types without bias, making them suitable when linkage specificity is unknown. The K63‑specific TUBE selectively binds K63 chains with ~20 nM affinity and can be combined with K48 TUBE HF or M1 TUBE for detailed linkage analysis. Magnetic‑TUBEs, which attach TUBE units to magnetic beads, allow rapid, one‑step pull‑down and downstream analysis of polyubiquitinated proteins by Western blotting or proteomics.
