Jacalin is a lectin composed of four subunits of approximately 16 kDa each that binds to O-glycosidically linked oligosaccharides. It preferentially binds the galactosyl (β-1,3) N-acetylgalactosamine structure, the T-antigen oligosaccharide to which peanut agglutinin (PNA) also binds. However, unlike PNA, Jacalin binds a mono- or disialylated form of this structure. Jacalin has been used to purify human IgA, and its specificity also affords the opportunity to localize or isolate glycoproteins with O-glycosidically linked oligosaccharide side chains. The recommended inhibiting/eluting sugar is 800 mM galactose or 100 mM melibiose.
