The Viability/Cytotoxicity Assay Kit for Animal Live & Dead Cells provides green/red fluorescent staining of live and dead cells, respectively. The assay employs two probes: Calcein AM that detects intracellular esterase activity and stains live cells green, and Ethidium homodimer III (EthD-III) that stains cells with compromised plasma membrane integrity red. The assay principal is applicable to most eukaryotic cell types, including adherent cells and certain tissues, but not bacteria or yeast, and can be used in place of trypan blue exclusion, 51Cr release, or similar methods for determining cell viability and cytotoxicity. The kit is suitable for fluorescence microscopy, fluorescence microplate reader, or flow cytometry. EthD-III, a superior alternative to the spectrally similar Ethidium homodimer I, has a higher DNA binding affinity with ~30% brighter fluorescence.Calcein (end-product of calcein AM hydrolysis): Ex/Em: 494/517 nm (pH 8); EthD-III: Ex/Em: 530/620 nm (with DNA).
