Gibco™ Viral Production Medium, AGT™
Description
- Gibco Viral Production Medium, AGT is the dry-format version of CTS Viral Production Medium, a chemically defined, serum-free, protein-free, animal origin-free, phenol red-free medium
- Medium is part of the CTS AAV-MAX Helper-Free AAV Production System for high-titer adeno-associated virus (AAV) production
- The CTS AAV-MAX Helper-Free AAV Production System is a scalable, high-titer platform for suspension AAV production with research-grade and GMP versions to enable seamless transition from research to commercial manufacturing
- Other components of the CTS AAV-MAX system include CTS Viral Production Cells 2.0, CTS AAV-MAX Transfection Kit, and CTS Viral-Plex Complexation Buffer.
Features of Viral Production Medium, AGT, include:
- Supports growth and transfection of CTS Viral Production Cells 2.0 from shake flask to bioreactor scales
- Helps enable production of high AAV titers, greater than 5 x 1010 vg/mL (unpurified)
- Simplifies transfer to manufacturing scale due to Advanced Granulation Technology (AGT) enabling auto-pH and simplified dissolution
- Formulated with GlutaMAX Supplement, no further supplementation required
- Manufactured in conformity with cGMP for medical devices, 21 CFR, Part 820, and certified to ISO 13485. Drug Master File and Regulatory Support File available upon request.
Advanced Granulation Technology
- Advanced Granulation Technology (AGT) helps simplify medium preparation at large scale with fast rehydration
- A just-add water protocol, and low dust generation, creates less pressure build-up during sterile filtration
- For more information on AGT media preparation at large scale, see Hydration and scale-up of AGT medium in HyPerforma and imPULSE Single-Use Mixers

Frequently Asked Questions (FAQs)
You can refer to page 9 of the AAV-MAX Helper-Free AAV Production System User Guide (https://assets.thermofisher.com/TFS-Assets/LSG/manuals/MAN0019619_AAV-MAX_Helper-Free_AAV_ProductionSystem_UG.pdf) for detailed Vi-CELL XR settings. We recommend diluting cell count samples with PBS to eliminate precipitate artifacts that may be created by interaction with the Vi-CELL reagents.
While the full formulation for Viral Production Medium, AGT is proprietary information that cannot be disclosed, we can confirm that the medium contains a surfactant and that no additional surfactant would need to be added to the medium to support your suspension CTS VPC2.0 in 2 L bench-top bioreactor to 50 L bioreactor.
Viral Production Medium, AGT is formulated to contain glucose at a concentration of 6 g/L.
Viral Production Medium, AGT powder needs to be reconstituted in deionized distilled water or Water For Injection (WFI) for Cell Culture (Cat. No. A12873) and then immediately sterilized by membrane filtration (positive pressure recommended). A low protein binding filter (such as PVDF) with a pore size of 0.2 µM is recommended.
GlutaMAX Supplement (Cat. No. 35050061) is already present in the one-part Viral Production Medium, AGT powder. Once rehydrated, the formulations of the two products are identical provided the research-grade Viral Production Medium (Cat No. A48179) has been supplemented with GlutaMAX Supplement at point of use.
