LifeSensors has developed monoclonal antibodies that detect both polyubiquitylated proteins and free ubiquitin. VU‑1 recognizes all ubiquitin linkages and provides strong, low‑background detection in Western blotting and immunostaining, enabling visualization of subcellular ubiquitin localization. Ubiquitin is an 8 kDa protein found in all eukaryotic cells and is attached to substrates through an enzymatic cascade involving E1, E2, and E3 enzymes. Its seven lysines allow formation of diverse polyubiquitin chains, with K48 and K63 being the best characterized. K48 chains typically target proteins for proteasomal degradation, while K63 chains regulate signaling, endocytosis, and immune responses. Research of less common linkage types remains ongoing.
