WEE1 kinase is crucial for the G2-M cell-cycle checkpoint, which is involved in DNA repair before mitotic entry. While normal cells repair damaged DNA during G1 arrest, cancer cells often have a deficient G1-S checkpoint and rely on a functional G2-M checkpoint for DNA repair. WEE1 is found to be overexpressed in various cancer types. WEE1-IN-3 inhibits cancer cell growth with IC50 values of 100-1000 nM for SW480 cells and less than 100 nM for H23 cells.
