Inhibit both GSK3β and GSK3α with CHIR99021. Together with other small molecules and growth factors, CHIR99021 maintains undifferentiated mouse ES cells (Ying et al. Nature, 2008), promotes self-renewal of human ES cells and mouse epiblast stem cells (Kim et al. Nat Commun, 2013), and allows for lineage reprogramming of fibroblasts to mature neurons (Hu et al. Cell Stem Cell, 2015; Li et al. Cell Stem Cell, 2009). This WNT activator also promotes the growth of mouse and human intestinal stem cells (Wang et al. Gastroenterology, 2013), and promotes the differentiation of human iPS cells to insulin-producing cells (Kunisada et al. Stem Cell Res, 2012) and cardiomyocytes (Lian et al. Nat Protoc, 2013).
