Description
- Peptide thiobenzyl esters are sensitive substrates for serine and cysteine proteases.
- The thiol leaving group is detected by reaction with a thiol reagent (DTNB or Leman's reagent) or 4,4'-dithiodipyridine.
- The rate of thiobenzyl ester hydrolysis is measured at 324 nm by monitoring the formation of 4-thiopyridone (E324nm = 19,800 M-1cm-1) or at 405 nm using DTNB as a thiol reagent (E405nm = 13,260 M-1cm-1) for 2-nitro-5-thiobenzoate anion TNBS.
