Lometrexol disodium is an antipurine antifolate that inhibits glycinamide ribonucleotide formyltransferase (GARFT) and human Serine hydroxymethyltransferase1/2 (hSHMT1/2). This leads to the inhibition of de novo purine synthesis, causing abnormal cell proliferation, apoptosis, and cell cycle arrest, thus exhibiting anticancer activity.
