OCT-3/4 (also known as POU5F1) is a transcription factor that has been recognized as fundamental in the maintenance of pluripotency in embryonic stem cells and primordial germ cells. It has been proposed as a useful marker for germ cell tumors (GCT) that exhibit features of pluripotentiality (seminoma/dysgerminoma, germinoma and embryonal carcinoma. OCT-3/4 immunostaining has been shown to be sensitive and specific for GCT, whether in primary gonadal or extragonadal sites or in metastatic lesions.

Expression of smooth muscle myosin is developmentally regulated, appearing early in smooth muscle development, and is specific for smooth muscle development. Two isoforms of smooth muscle myosin heavy chain have been identified, designated MHC-1 and MHC-2, they are differentially expressed depending on the functional activity of the muscle.\nThe antibody may be useful for the study of breast tumors as the presence of an intact layer of myoepithelial cells is an important feature, which may distinguish benign breast lesions and carcinoma in situ from invasive tumors

Recognizes a protein of 48kDa, which is identified as Adipophilin. It belongs to the perilipin family, members of which coat intracellular lipid storage droplets. This protein is associated with the lipid globule surface membrane material, and maybe involved in development and maintenance of adipose tissue. However, it is not restricted to adipocytes as previously thought, but is found in a wide range of cultured cell lines, including fibroblasts, endothelial and epithelial cells, and tissues, such as lactating mammary gland, adrenal cortex, Sertoli and Leydig cells, and hepatocytes in alcoholic liver cirrhosis, suggesting that it may serve as a marker of lipid accumulation in diverse cell types and diseases.

Diagnostic Biosystems P504S p504S/Alpha-Methylacyl-CoA Racemase (AMACR)
P504S (AMACR) is an essential enzyme in the β-oxidation of branched-chain fatty acids. Recently, P504S was identified through cDNA library subtraction and microarrays in malignant prostate tissues. High expression of P504S protein is found in prostatic adenocarcinoma but not in benign prostatic tissue by immunohistochemical staining in paraffin-embedded tissues. The expression of P504S is also detected in two premalignant lesions of the prostate: high-grade prostatic intraepithelial neoplasia (PIN) and atypical adenomatous hyperplasia. Using P504S as a positive marker along with basal cell staining (34 βE12 or P63) as a negative marker could help to confirm the diagnosis of small focus of prostate carcinoma on needle biopsy.

Electron Microscopy Sciences Microwave Incubation Tray for Immunostaining
For immunostaining, we recommend a water load that is the same size as the bottom of the microwave cavity in order not to disturb the distribution of microwave energy within the cavity. In our research, we found that between 150 and 200 ml of water in the bottom of the incubation tray prevents evaporation of the droplets of reagent. A PAP pen should be used to maintain the droplets at a uniform size and shape.
