Mutations in this gene are a cause of Charcot-Marie-Tooth disease type 4D, and expression of this gene may be a prognostic indicator for several types of cancer. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene.

Chemokines form a superfamily of secreted proteins involved in immunoregulatory and inflammatory processes. The superfamily is divided into four subfamilies based on the arrangement of the N-terminal cysteine residues of the mature peptide. This chemokine, a member of the CC subfamily, is secreted by activated T cells and displays chemotactic activity for monocytes but not for neutrophils. It binds to the chemokine (C-C motif) receptor 8.

Myosin phosphatase target subunit 1 (MBS) is a subunit of myosin phosphatase, which regulates the actin-myosin interaction downstream of the GTPase Rho. RhoA, in its active GTP-bound form, interacts with MBS to control myosin light chain (MLC) phosphorylation, affecting smooth muscle contraction and nonmuscle cell function. RhoA activates Rho-kinase, which phosphorylates MBS and inactivates myosin phosphatase. Overexpression or activation of RhoA increases MBS and MLC phosphorylation. This pathway regulates the balance of actin and myosin interaction. Several isoforms of this gene have been identified due to alternative splicing.
