Gel Loading Dye, Orange (6X) is a pre-mixed loading buffer with a tracking dye for agarose and non-denaturing polyacrylamide gel electrophoresis. This solution contains SDS, which often results in sharper bands, as some restriction enzymes are known to remain bound to their DNA substrates following cleavage. EDTA is also included to chelate magnesium (up to 10 mM) in enzymatic reactions, thereby stopping the reaction. Orange G will not appear in gel photographs, and runs ahead of all but the smallest restriction fragments. Orange G migrates at approximately 50 bp on a standard 1% TBE agarose gel. GelGreen is a sensitive, stable and environmentally safer alternative to the highly toxic ethidium bromide (EB) for staining dsDNA, ssDNA or RNA in agarose or polyacrylamide gels. The dye is noncytotoxic, nonmutagenic and nonhazardous at concentrations used in gel staining. It can be conveniently disposed down the drain or in regular trash at working concentrations, reducing disposal costs. It is much more sensitive than EB or SYBR Safe and extremely stable for long-term room temperature storage. Simple to use in precast or post-electrophoresis gel staining. Compatible with a UV transilluminator, gel reader or scanner with visible light, blue LED, or 488 nm laser-based excitation; replaces SYBR with no optical setting change. This stain contains a phospho-tag™ group, which allows direct, in-gel detection of phosphate groups attached to tyrosine, serine, or threonine residues, without the need for antibodies or radioisotopes. The stain can be used with standard SDS-polyacrylamide gels or with 2-D gels.
