Reverse transcription polymerase chain reaction (RT-PCR) is a laboratory technique that combines the reverse transcription of RNA into DNA and the subsequent amplification of the DNA using PCR.
In RT-PCR, a reverse transcriptase enzyme converts an RNA template to its complementary DNA (cDNA). The cDNA then becomes the template for PCR amplification.
A typical RT-PCR kit includes the main components needed for the assay:
- Buffer
- Primers – a combination of deoxynucleotide triphosphates (dNTPs) of the four bases of adenine (dATP), cytosine (dCTP), guanine (dGTP), and thymine (dTTP)
- Reverse transcriptase reagent
The combination of RT-PCR and qPCR techniques (sometimes labelled RT-qPCR) is often used in research and clinical labs to analyze gene expression and quantify viral RNA. Since this method uses fluorescence to track the amplification reaction, these kits may also include:
- Fluorogenic primers or labeled fluorogenic probes
- Fluorescent dye
Reagents for cDNA PCR can be purchased separately or as cDNA synthesis kits.
RT-PCR is commonly used for RNA detection, molecular cloning, and gene sequencing. RT-qPCR is used for gene expression analysis, RNAi or microarray validation, pathogen detection, genetic testing, and other disease research.

Lyo-Ready qPCR Mix, 2.6x is a higher concentration, glycerol-free formulation that contains Taq polymerase, reaction buffer, dNTP, MgCl2 and lyo-excipients. It is ideal for developing room temperature, lyophilized, multiplex qPCR diagnostic tests and is suited for use with microfluidic qPCR, where the initial volume needs to be smaller. It can be used as a wet mix or it can be lyophilized without impacting its performance post rehydration.

Meridian’s RT-qPCR Extraction Control Orange uses artificial micelles that contain quantitative PCR positive control RNA sequence (with no known homology to any organism, so that it does not interfere with the detection of the sample target RNA). The cells are added to the RT-qPCR Extraction Control is spiked in the with lysis buffer into the sample prior to RNA extraction for downstream amplification by RT-qPCR. The detection of RT-qPCR Extraction Control acts as a cDNA synthesis and PCR positive control and confirms the success of the extraction and amplification steps, avoiding the misinterpretation of a false negative result.

Meridian Life Science Low DNA qPCR Mix, 5ML
Low DNA qPCR master mix contains a chemical hot-start Taq polymerase and highly optimized reaction buffer with dNTPs, MgCl2, stabilizers and PCR enhancers. The Taq polymerase is purified twice, before and after the addition of the chemical hot-start, this helps to remove trace amounts of DNA (known as bioburden) that come from the manufacturing of the DNA polymerase. In addition there is no bioburden that comes with antibody hot-start, making it ideal for low-copy number targets and also microbial DNA and fungal DNA detection assays. Low DNA qPCR master mix can also be used for fast, precise and highly reproducible genotyping of sequence variants, including loci, provides higher stringency and more specific binding of the allele-specific probes, with wider and clearer separation of allele cluster.

BioChain's HotStart Taq Polymerase combines high specificity, sensitivity, and minimal optimization with a fast 10-minute activation time. During PCR reaction setup, DNA polymerases commonly exhibit some synthesis activity which can lead to amplification of mis-primed sequences and accumulation of primer-dimers that compromise the specificity and yield of the reaction. Non-specific amplification can be minimized with HotStart Taq Polymerase. Our ready-to-use HotStart Taq Polymerase is chemically modified causing its activity to be blocked at lower temperatures. Activity is restored when the enzyme is heated to 95oC during the initial denaturing process, enabling hot-start PCR.
