Universal Pre-enrichment Broth is used for the recovery of Salmonella spp. and Listeria spp. in a laboratory setting. Universal Pre-enrichment Broth is not intended for use in the diagnosis of disease or other conditions in humans. Traditional methods for recovering Salmonella spp. and Listeria spp. from food products require separate pre-enrichment media for each microorganism. Some broth media recommended for preenrichment contain antibiotic inhibitors or have insufficient buffering capacity which hinder recovery of sublethally injured cells. Bailey and Cox formulated Universal Preenrichment Broth to permit simultaneous resuscitation of sublethally injured Salmonella and Listeria. Buffered Peptone Water (BPW) is used for the non-selective pre-enrichment of Salmonella spp. from food and is not intended for use in the diagnosis of disease or other conditions in humans. Edel and Kamelmacher found that food preservation techniques involving heat, desiccation, preservatives, high osmotic pressure, or pH changes cause sublethal injury to Salmonella spp. Preenrichment in a non-selective medium allows for repair of cell damage and facilitates the recovery of Salmonella. Lactose Broth is frequently used for this purpose, but it may be detrimental to recovering Salmonellae. Buffered Peptone Water maintains a high pH over the pre-enrichment period and allows in repair of injured cells that may be sensitive to low pH. This is particularly important for vegetable specimens which have a low buffering capacity. Buffered Peptone Water is used in standard methods. Eosin Methylene Blue Agar Levine is used for the isolation and differentiation of Gram-negative enteric bacilli and is not intended for use in the diagnosis of disease or other conditions in humans.This medium was introduced in 1916 by Holt-Harris and Teague to differentiate Escherichia spp. and Aerobacter spp. It was modified by Levine in 1918 who removed sucrose from the formula and increased the lactose content. The distinctive metallic sheen produced by E. coli on this medium is due to acid production resulting in an amide bonding between the eosin and methylene blue, other coliforms do not produce enough acid to cause this reaction.
