A-1 Medium is used for the detection of coliform organisms in water and food in a laboratory setting. A-1 Medium is not intended for use in the diagnosis of disease or other conditions in humans.Since the early 1900’s enumeration of coliform organisms, specifically E. coli, have been used to determine water purity. Elevated temperature and most-probable-number (MPN) methods are routinely used for analysis of water and food samples for presence of fecal coliforms. A-1 Medium was formulated to hasten E. coli recovery and reduce the incidence of false positive cultures.In 1972 Andrews and Presnell developed A-1 Medium. This medium recovers E. coli from estuarine water in 24 hours instead of 72 hours, and in greater numbers without the pre-enrichment step. Modified Semi-Solid Rappaport-Vassiliadis (MSRV) Agar is used with Novobiocin for the rapid detection of motile Salmonella spp. and is not intended for use in the diagnosis of disease or other conditions in humans. MSRV was developed in 1986 by De Smedt, Bolderdijk and Rappold as a rapid means of Salmonella detection. The medium, based upon Rappaport Vassiliadis broth, is inoculated directly from the preenrichment medium, in the center of the plate. Motile organisms spread from the center in the semi-solid agar, but non-salmonellas are inhibited by the selective agents. Universal Pre-enrichment Broth is used for the recovery of Salmonella spp. and Listeria spp. in a laboratory setting. Universal Pre-enrichment Broth is not intended for use in the diagnosis of disease or other conditions in humans. Traditional methods for recovering Salmonella spp. and Listeria spp. from food products require separate pre-enrichment media for each microorganism. Some broth media recommended for preenrichment contain antibiotic inhibitors or have insufficient buffering capacity which hinder recovery of sublethally injured cells. Bailey and Cox formulated Universal Preenrichment Broth to permit simultaneous resuscitation of sublethally injured Salmonella and Listeria.
