This MAb recognizes basic (Type II or HMW) cytokeratins, which include 67 kDa (CK1); 64 kDa (CK3); 59 kDa (CK4); 58 kDa (CK5); 56 kDa (CK6); 52 kDa (CK8). Twenty human keratins are resolved with two-dimensional gel electrophoresis into acidic (pI 6.0) subfamilies. The acidic keratins have molecular weights (MW) of 56.5, 55, 51, 50, 50 , 48, 46, 45, and 40 kDa. MAb AE3 recognizes the 65-67, 64, 59, 58, 56, and 52 kDa keratins of basic subfamily. Many studies have shown the usefulness of keratins as markers in cancer research and tumor diagnosis. Recognizes a protein of 75 kDa, identified as γ heavy chain of human immunoglobulins. It reacts with all sub-classes of γ chain of human immunoglobulins. It does not cross-react with α (IgA), μ (IgM), ε (IgE), or δ (IgD), heavy chains, T-cells, monocytes, granulocytes, or erythrocytes. This MAb is useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. The most common feature of these malignancies is the restricted expression of a single heavy chain class. This MAb is specific for progesterone. It exhibits minimal cross reactivity with related compounds in ELISA. It reacts with Progesterone-11a-HMS-BSA: 100%; 5-beta-Pregnane-3,20-dione: 48%; 5-alpha-Pregnane-3,20-dione: 26.4%; 17-alpha-Hydroxyprogesterone: 2.5% and 20-alpha-Hydroxyprogesterone: 0.04%. This antibody recognizes the Calcineurin A alpha protein, encoded by the PPP3CA (Protein Phosphatase 3 Catalytic Subunit Alpha) gene. Calcineurin is an enzyme that dephosphorylates serine and threonine residues in proteins. It is a heterodimer of a 59 kDa catalytic A subunit and a 19 kDa regulatory B subunit that is activated by the binding of calcium ions and calmodulin. Calcineurin is expressed in many tissues, but its levels are highest in the brain, where it may play a role in learning and memory. It has many substrates, including NFAT, a transcription factor that is activated by dephosphorylation. Complexes of the immuno-suppressants cyclosporine and FK506 with immunophilin proteins such as cyclophilin and FKBP12 are potent and specific inhibitors of Calcineurin activity. This antibody recognizes a protein of 21 kDa, identified as the Bax protein. This MAb is highly specific to Bax and shows no cross-reaction with Bcl-2 or Bcl-X protein. Bcl-2 blocks cell death following a variety of stimuli. Bax has extensive amino acid homology with Bcl-2 and it homodimerizes and forms heterodimers with Bcl-2. This MAb recognizes a protein of ~50 kDa which is identified as Glial Fibrillary Acidic Protein (GFAP). It shows no cross-reaction with other intermediate filament proteins. GFAP is specifically found in astroglia. GFAP is a very popular marker for localizing benign astrocyte and neoplastic cells of glial origin in the central nervous system.
