This MAb reacts with a 58 kDa protein identified as vimentin. It shows no cross-reaction with other closely related intermediate filament proteins (IFP s) such as desmin, keratin, neurofilament, and glial fibrillary acid protein.Anti-vimentin alone is of limited value as a diagnostic tool; however, when used in panels with other antibodies, it is useful for the sub-classification of a given tumor. Expression of vimentin, when used in conjunction with anti-keratin, is helpful when distinguishing melanomas from undifferentiated carcinomas and large cell lymphomas. All melanomas and Schwannomas react strongly with anti-vimentin. It labels a variety of mesenchymal cells, including melanocytes, lymphocytes, endothelial cells, and fibroblasts. Non-reactivity of anti-vimentin is often considered more useful than its positive reactivity, since there are a few tumors that do not contain vimentin, e.g. hepatoma and seminoma. Anti-vimentin is also useful as a tissue process control reagent.P

This antibody recognizes a 32 kDa protein, which is identified as homodimer of galectin-13 (also known as PP13). Galectins are a family of soluble beta-galactoside-binding lectins that modulate cell-to-cell adhesion and cell-to-extracellular matrix (ECM) interactions and play a role in tumor progression, pre-mRNA splicing and apoptosis. One such member, galectin-13, is a 139 amino acid protein that contains one galectin domain. Possessing lysophospholipase activity, galectin-13 exists as a disulfide-linked homodimer. Galectin-13 is suggested to have a developmental role in the placenta and may display immunobiological activity in feto-maternal blood-spaces. This antibody recognizes an intermediate filament protein (IFP) of 55 kDa, which is identified as cytokeratin 7. This MAb is highly specific to cytokeratin 7 and shows no cross-reaction with other IFPs. Cytokeratin 7 is a basic cytokeratin, which is found in most glandular and transitional epithelia but not in the stratified squamous epithelia. Keratin 7 is expressed in the epithelial cells of ovary, lung, and breast but not of colon, prostate, or gastrointestinal tract. CD6 is a type I transmembrane glycoprotein that contains a 24-amino acid signal sequence, three extracellular scavenger receptor cysteine-rich (SRCR) domains, a membrane-spanning domain and a 44-amino acid cytoplasmic domain. The CD6 glycoprotein is tyrosine phosphorylated during TCR-mediated T cell activation. CD6 shows significant homology to CD5. CD6 is present on mature thymocytes, peripheral T cells and a subset of B cells. This antibody recognizes a protein of 31 kDa, which is identified as MTAP (5'-deoxy-5'-methylthioadenosine phosphorylase). It catalyzes the reversible phosphorolysis of methylthioadenosine, which is important in polyamine metabolism and for the salvage of adenine and methionine. The gene encoding MTAP is linked to the tumor suppressor gene, p16INK4A. Deficient levels of MTAP can occur in cancers primarily through co-deletion of the MTAP gene and the p16INK4A gene. This MAb recognizes a protein of 56.5 kDa, identified as cytokeratin 10 (CK10). CK10 is expressed in all suprabasal layers of the epidermis. In the epidermis, expression of CK10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. However, CK10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), CK10 is detected very frequently. This antibody recognizes a protein of 75 kDa, identified as Thrombomodulin. Thrombomodulin is a transmembrane glycoprotein with natural anticoagulant properties. It is normally expressed by a restricted number of cells, such as endothelial and mesothelial cells. In addition, synovial lining and syncytio-trophoblasts of placenta also express thrombomodulin. This protein is present in almost all of benign vascular tumors and majority of malignant vascular tumors (Kaposi's sarcoma, angiosarcoma, and epithelioid hemangioendothelioma). Hence, anti-thrombomodulin serves as a sensitive marker for lymphatic endothelial cells and their tumors.
