Tryptic Soy Broth (TSB), also known as Soybean-Casein Digest (SCD) Medium, is a general nutrient and enrichment broth. It is used in qualitative procedures for the cultivation of fastidious and non-fastidious microorganisms and is also recommended for sterility testing. TSB is recommended for use as the inoculum broth for disc diffusion and agar dilution antimicrobial susceptibility testing by the CLSI. Originally developed in 1960 by de Man, Rogosa & Sharpe, the medium is suitable for most lactic acid bacteria and is intended as a substitute for Tomato Juice Agar. Selectivity against Streptococci and Fungi is provided by ammonium citrate and sodium acetate. Congo red is utilized to stain the cells of fungi and the outer membrane of Gram-negative bacteria. Additionally, studies have indicated virulent Shigella flexneri serotype 2a, grown in the presence of congo red at 37°C, showed increased levels of three proteins with MRS of 43K, 58K, and 63K proteins) in the Sarkosyl-soluble membrane fractions. Oral Treponeme Enrichment Broth (OTEB) is an enriched nonselective medium used for the isolation, cultivation, and maintenance of Treponema spp. from clinical specimens. This medium is prepared, dispensed, and packaged under oxygen-free conditions to prevent the formation of oxidized products prior to use.OTEB is a complex peptone yeast extract medium containing volatile fatty acids and calf serum. Volatile fatty acids and serum are necessary growth factors for the recovery of Treponemes from clinical specimens. A variety of sugars have been added as a source of carbohydrates. The addition of short-chain fatty acids (SCFA) and serum may also aid in the recovery and growth of fastideous organisms with complex nutritional requirements.
