Vector Biolabs aryl hydrocarbon receptor nuclear translocator Adenovirus (Ad-HIF1b/ARNT)
The aryl hydrocarbon (Ah) receptor is involved in the induction of several enzymes that participate in xenobiotic metabolism. The ligand-free, cytosolic form of the Ah receptor is complexed to heat shock protein 90. Binding of ligand, which includes dioxin and polycyclic aromatic hydrocarbons, results in translocation of the ligand-binding subunit only to the nucleus. Induction of enzymes involved in xenobiotic metabolism occurs through binding of the ligand-bound Ah receptor to xenobiotic responsive elements in the promoters of genes for these enzymes. This gene encodes a protein that forms a complex with the ligand-bound Ah receptor, and is required for receptor function. The encoded protein has also been identified as the beta subunit of a heterodimeric transcription factor, hypoxia-inducible factor 1 (HIF1)

pSNAPf Vector is a mammalian expression plasmid intended for the cloning and stable or transient expression of SNAP-tag protein fusions in mammalian cells. This plasmid encodes SNAPf, a SNAP-tag protein, which is expressed under control of the CMV promoter. The expression vector has an IRES (internal ribosome entry site) and a neomycin resistance gene downstream of the SNAPf for the efficient selection of stable transfectants. pSNAPf Vector contains two multiple cloning sites to allow cloning of the fusion partner as a fusion to the N- or C-terminus of the SNAPf. The SNAP-tag is a novel tool for protein research, allowing the specific, covalent attachment of virtually any molecule to a protein of interest. The SNAP-tag is a small protein based on mammalian O6-alkylguanine-DNA-alkyltransferase (AGT). SNAP-tag substrates are derivatives of benzyl purines and benzyl pyrimidines. In the labeling reaction, the substituted benzyl group of the substrate is covalently attached to the SNAP-tag.

The pESG-IBA102 vector is designed for high-level, stable, and non-replicative transient expression of target proteins with a C-terminal Twin-Strep-tag in mammalian cells. The vector carries an ampicillin resistance cassette for selection of transformed E. coli cells as well as the F1 and ColE1 origin for a high plasmid copy number. In addition, it carries the human cytomegalovirus (CMV) immediate-early promoter for high-level expression, the neomycin resistance gene for selection of stable cell lines, and the SV40 ori for episomal replication in cell lines that are latently infected with SV40 or that express the SV40 large T antigen (e.g. HEK293T, COS-1, COS-7). BM40 secretory signal peptide is encoded for the transfer of the expressed protein into the medium. During translocation from the cytosol the signal peptide is removed from the protein. In addition to the direct cloning of the gene of interest into pESG-IBA vectors with Esp3I, another option via Entry Vector is possible.

The protein encoded by this gene is a member of the serine/threonine kinase family. In response to cellular stress and proinflammatory cytokines, this kinase is activated through its phosphorylation by MAP kinases including MAPK1/ERK, MAPK14/p38-alpha, and MAPK11/p38-beta. In vitro, this kinase phosphorylates heat shock protein HSP27 at its physiologically relevant sites.

Programmed cell death, known as apoptosis, of nonessential cells is necessary for embryogenesis, metamorphosis, tissue turnover and proper development and function of the immune system. Apoptosis causes cytoplasmic condensation, nuclear fragmentation and membrane blebbing. There are several proteins that are responsible for the balance of signals that confer cell death and/or cell survival. Among these are the proteins of the tumor necrosis factor ligand superfamily, which includes FAS (APO-1, CD95), FAS ligand (APO-1L, CD178), TRAIL and TWEAK (APO3L) and the FAS accessory protein FAF1 (FAS-associated protein factor-1).

The breast cancer susceptibility gene, BRCA1, which maps to chromosome 17q is expressed in numerous tissues, including breast and ovary. A second breast cancer susceptibility gene, BRCA2, located on chromosome 13q12-13, also confers a high incidence of breast and ovarian cancer. Mutations within the BRCA1 gene may account for at least 80% of families with increased incidence of both early-onset breast cancer and ovarian cancer.This adenovirus expresses the human BRCA1 transcript variant 5 (NM_007299). This variant differs in the 5' UTR, uses two alternate in-frame splice sites in the central coding region, and lacks an alternate exon in the 3' coding region that results in a frameshift, compared to variant 1.
