Biotium MemBrite™ Fix 640/660 Cell Surface Staining Kit, 500 Reactions
MemBrite™ Fix Cell Surface Staining Kits are designed for covalently staining the surface of live cells to conveniently visualize cell boundaries in immunofluorescence experiments. MemBrite™ Fix are new covalent cell surface stains that react irreversibly with cell surface proteins for staining that can withstand formaldehyde or alcohol fixation, and detergent permeabilization. Unlike lectins such as WGA, which bind specific targets that may vary between cell types, MemBrite™ Fix dyes react widely with cell surface proteins. MemBrite™ Fix staining is rapid and non-toxic to cells, and because MemBrite™ Fix dyes are highly water soluble, they stain cells much more evenly than traditional lipophilic membrane dyes like DiO, DiI, PKH, Vybrant, or CellMask™. This kit has reagents sufficient for 500 labeling reactions. The kit contains 5 vials of MemBrite™ Fix 640/660. Each vial is sufficient for 20 uL of 1000X dye after DMSO reconstitution.
Biotium Human Transferrin, CF568 Conjugate, 1 mg
Transferrin is a glycoprotein that binds iron for delivery to vertebrate cells through receptor-mediated endocytosis. After binding to its receptor on the cell surface, transferrin is rapidly internalized by invagination of clathrin-coated pits with formation of endocytic vesicles, the acidic environment favors dissociation of iron from the transferrin-receptor complex. Following the release of iron, the apotransferrin is recycled to the plasma membrane where it is released from its receptor to scavenge more iron. Fluorescent conjugates of human transferrin can be used as tracers for investigation of endocytosis by microscopy. CF dyes are Biotium's line of next-generation fluorescent dyes with advantages in brightness, photostability, and conjugate specificity compared to other fluorescent dyes. Red fluorescent CF568 dye has excitation/emission at 562/583 nm.

AdipoGen Sulfo-NHS-LC-Biotin (50 mg)
Chemical. CAS: 191671-46-2127062-22-0. Formula: C20H29N4NaO9S2. MW: 556.59. Sulfo-NHS-LC-Biotin is an amino-reactive biotinylation reagent with a long spacer arm. The 6-aminocaproic acid spacer arm provides distance between the biotin and the target ligand which helps alleviate steric hindrance, in addition it forms permanent amide bonds. The sodium sulfonate groups render this compound water-soluble and prevent it from crossing the lipid bi-layer of the cell membrane, making it a tool to label cell surface proteins. In addition, Sulfo-NHS-LC-Biotin is used to biotinylate antibodies, proteins, peptides, or amino acids by reacting with primary amines as found in the N-terminus and Lysine side chains. Optimal reaction conditions are at a pH of 7-9. Avoid amine-containing buffers, which may compete in the acylation reaction.

Peanut agglutinin (PNA) binds the T-antigen, a galactosyl (β-1,3) N-acetylgalactosamine structure present in many glycoconjugates, such as M and N blood groups, gangliosides, and other soluble and membrane-associated glycoproteins and glycolipids. With exceptions, the PNA receptor sequence is normally sialylated, which prevents binding to its receptor oligosaccharide (see Jacalin). Sialic acid not bound to receptor sugars may also inhibit binding, and including calcium in diluents can enhance binding. This fluorescein-labeled PNA features a ratio of fluorophores to lectin protein that provides optimal staining (excitation 495 nm, emission 515 nm). Supplied as a solution essentially free of unconjugated fluorophores, it is preserved with sodium azide. The recommended inhibiting/eluting sugar is 200 mM galactose.
