Recognizes a protein of 75 kDa, identified as γ heavy chain of human immunoglobulins. Its epitope maps in CH2 domain of Fc region of IgG. It reacts with all sub-classes of γ chain of human immunoglobulins. It does not cross-react with α (IgA), μ (IgM), ε (IgE), or δ (IgD), heavy chains, T-cells, monocytes, granulocytes, or erythrocytes. Reacts with five invariable CD3 chains designated CD3y, CD3d, CD3e, CD3f, and CD3n with molecular weight ranging from 16-28 kDa. CD3, also known as T3, is a member of the immunoglobulin superfamily that plays a role in antigen recognition, signal transduction, and T cell activation. It is found on all mature T-lymphocytes, NK, T-cells, and some thymocytes. It is expressed, typically at high levels, on peripheral T cells and majority of T cell neoplasms. Thymocytes express CD3 at different level on the cell surface in the course of differentiation and, in cortical thymus, CD3 is predominantly intracytoplasmic. Mucosa associated lymphoid tissue lymphoma translocation gene 1 (MALT1) is found in extranodal low-grade B cell lymphomas. MALT1 encodes two Ig-like C2-type domains and fuses with an API2 gene, which is highly expressed in adult lymphoid tissue. The translocation of this MALT1 gene, which maps to human chromosome 18q21, and the apoptosis-inhibiting API2 gene results in an increased development of MALT lymphomas and apoptosis inhibition. Sites at which this API2-MALT1 (11;18)(q21;q21) translocation commonly occurs are within human lung and kidney tissue. MALT lymphoma expresses nuclear Bcl10, which mediates the oligomerization and activation of a MALT1 caspase-like domain. Recognizes a cell surface glycoprotein of 80-95 kDa (CD44) on lymphocytes, monocytes, and granulocytes (Leucocyte Typing Workshop V). Its epitope is resistant to digestion by trypsin and chymotrypsin. The CD44 family of glycoproteins exists in a number of variant isoforms, the most common being the standard 85-95 kDa or hematopoietic variant (CD44s). Higher molecular weight isoforms are described in epithelial cells (CD44v), which are believed to function in intercellular adhesion and stromal binding. Ten to forty percent of the patients with acquired immunodeficiency syndrome (AIDS) develop cytomegalovirus (CMV) infections. In some patients with AIDS, CMV is detected in the bronchoalveolar lavage fluid (BALF), urine, and other specimens, even when there are no symptoms of CMV disease. An indicator of active CMV infection is needed to facilitate the diagnosis of CMV disease in patients with AIDS or HIV infection. Bcl-x belongs to the Bcl-2 family of proteins and plays a crucial role in apoptosis. Bcl-X has two isoforms: Bcl-XL (long), a 26 kDa protein which suppresses cell death, and Bcl-XS (short), a 178 amino acid protein lacking a 63 amino acid domain which functions as a dominant inhibitor of Bcl-2 and promotes apoptosis. This antibody recognizes a protein of 25-26 kDa, identified as Bcl-2. This antibody shows no cross-reaction with Bcl-x or Bax proteins. Bcl-2 suppresses apoptosis in a variety of cell systems including factor-dependent lymphohematopoietic and neural cells. It regulates cell death by controlling the mitochondrial membrane permeability. Bcl-2 appears to function in a feedback loop system with caspases. It inhibits caspase activity either by preventing the release of cytochrome c from the mitochondria and/or by binding to the apoptosis-activating factor (APAF-1). Expression of Bcl-2 inhibits programmed cell death (apoptosis). In most follicular lymphomas, neoplastic germinal centers express high levels of Bcl-2 protein, whereas the normal or hyperplastic germinal centers are negative. This MAb reacts with a protein of 22 kDa, identified as β sub-unit of HCG. It does not cross react with the α sub-unit. HCG is a glycoprotein, which is secreted in large quantities by normal trophoblasts. It is present only in trace amounts in non-pregnant urine and sera but rises sharply during pregnancy. HCG is composed of two non-identical, non-covalently linked polypeptide chains designated as the alpha and beta subunits. The alpha subunit is identical to that of thyroid stimulating hormone (TSH), follicle stimulating hormone (FSH), and luteinizing hormone (LH). hCG MAb detects cells and tumors of trophoblastic origin such as choriocarcinoma. Recognizes a protein of 210-220 kDa, which is identified as the complement receptor 1 (CR1)/CD35. This MAb is specific for a site in CR1 that is distal from the C3b-binding site, so that it is unable to block CR1 activity. This MAb is highly specific to CR1 and shows no cross-reaction with CR2. The primary function of CR1 is to serve as the cellular receptor for C3b and C4b, the most important components of the complement system leading to clearance of foreign macromolecules. Recognizes a protein of 33-37 kDa, identified as CD20 (Workshop V; Code CD20.12). B9E9 recognizes extracellular domain of CD20. The epitope is similar to or identical to that recognized by other CD20 antibodies including Leu-16 and B1. This MAb can be used for immunophenotyping of leukemia and malignant cells, B lymphocyte detection in peripheral blood, B cell localization in tissues and B lymphocyte purification by immunosorbent methods.
