The SensoLyte Plus 520 MMP-9 Assay Kit is designed for specifically detecting MMP-9 activity in biological samples which may contain multiple MMPs, such as culture medium, serum, plasma, synovial fluid, and tissue homogenate. A specific anti-MMP-9 monoclonal antibody is used in combination with a MMP fluorogenic substrate, 5-FAM/QXL520 FRET peptide. The fluorescence signal is monitored at Ex/Em=490 nm/520 nm upon MMP-9-induced cleavage of the FRET substrate. Ample materials are provided to perform 96 assays in a 96-well format.

Its actions can result in protein processing, activation or degradation. Matriptase is widely expressed in virtually all epithelium and is specifically found in tumors of epithelial origin. Matriptase has been implicated in carcinogenesis including ovarian, prostate and cervical cancers and is considered a drug target for developing cancer therapeutic agent (1-3). This kit provides a convenient assay for screening of enzyme inhibitors and activators or for continuous assay of enzyme activity using a fluorogenic substrate. (excitation/emission=496 nm/520 nm)

The kit employs a simple enzymatic recycling reaction for total glutathione quantification. The kit has a detection sensitivity limit of 8 nM. Each kit provides sufficient reagents to perform up to 100 assays, including standard curve and unknown samples.
