The RosetteSep™ Human Total Lymphocyte Enrichment Cocktail is designed to enrich lymphocytes from whole blood by negative selection. Unwanted cells are targeted for removal with Tetrameric Antibody Complexes recognizing CD16, CD36, CD66b and glycophorin A on red blood cells (RBCs). When centrifuged over a buoyant density medium such as RosetteSep™ DM-L (Catalog #15705) or Lymphoprep™ (Catalog #07801), the unwanted cells pellet along with the RBCs. The purified lymphocytes are present as a highly enriched population at the interface between the plasma and the buoyant density medium.

Desired cells are targeted with Tetrameric Antibody Complexes recognizing CD19, CD20 and dextran-coated magnetic particles. Labeled cells are separated using an EasySep™ magnet without the use of columns. Cells of interest remain in the tube while unwanted cells are poured off. The CD19 antigen is present on mature B cells and B cell progenitors, but is lost upon maturation to plasma cells. The antigen CD20 is expressed on all stages of B cell development except on pro B cells or plasma cells.This product replaces the EasySep™ HLA Whole Blood B Cell Positive Selection Kit (Catalog #18184HLA) for even faster cell isolations.

Unwanted cells are targeted for removal with antibody complexes recognizing CD3, CD11b, CD19 and glycophorin A on red blood cells (RBCs). When centrifuged over a buoyant density medium such as Lymphoprep™ (Catalog #07801), the unwanted cells pellet along with the RBCs. The hematopoietic progenitor cells are present as an enriched population at the interface between the plasma and the buoyant density medium. Cells of interest can then be positively selected from the pre-enriched cell population using EasySep™ (e.g. EasySep™ Human CD34 Positive Selection).
