Complement C1q B Chain (C1qb) is a subunit of C1q. C1q is composed of 6 polypeptide chains each of C1qa, C1qb, and C1qc. C1q is a component of the classical complement pathway, and thereby plays an important role in the immune response. Each C1q subunit contains an N-terminal collagen-like region and a C-terminal C1q globular domain. The presence of receptors for C1q on effector cells modulates its activity, which may be antibody-dependent or independent. Macrophages are the primary source of C1q, while anti-inflammatory drugs as well as cytokines differentially regulate expression of the mRNA as well as the protein. Annexin A1 belongs to the annexin family. It plays a role in glucocorticoid-mediated down-regulation of the early phase of the inflammatory response. In resting conditions Annexin A1 is localized to the cytoplasm. Upon activation it is mobilized to the membrane and secreted. Annexin A1 and contains 4 annexin repeats. A pair of annexin repeats may form one binding site for calcium and a phospholipid. Expression of the ANXA1 gene is upregulated in hairy cell leukemia (HCL). Detection of ANXA1 provides a simple, highly sensitive and specific assay for diagnosing HCL. Annexin A1 has also been found to be protective against DNA damage induced by heat in breast cancer cells, suggesting it is involved in tumor suppressive and protective activities, and also is associated with treatment resistance. Aldo-keto Reductase Family 1 Member C2 (AKR1C2), also known as Dihydrodiol Dehydrogenase 2 (DDH2), is a member of the aldo/keto reductase superfamily, which consists of more than 40 known enzymes and proteins. These enzymes catalyze the conversion of aldehydes and ketones to their corresponding alcohols by utilizing NADH and/or NADPH as cofactors. The enzymes display overlapping but distinct substrate specificity. This enzyme catalyzes the reaction of progesterone to the inactive form 20-alpha-hydroxy-progesterone. This MAb recognizes a protein of ~30 kDa, identified as CELA3B (Chymotrypsin like elastase family member 3B). Elastases form a subfamily of serine proteases that hydrolyze many proteins in addition to elastin. Humans have six elastase genes which encode the structurally similar proteins elastase 1, 2, 2A, 2B, 3A, and 3B. Unlike other elastases, elastase 3B has little elastolytic activity. Like most of the human elastases, elastase 3B is secreted from the pancreas as a zymogen and, like other serine proteases such as trypsin, chymotrypsin and kallikrein; it has a digestive function in the intestine. Elastase 3B preferentially cleaves proteins after alanine residues. Elastase 3B may also function in the intestinal transport and metabolism of cholesterol. Recognizes a protein of 130 kDa, identified as Desmoglein-3 (DSG3). This MAb is highly specific to Desmoglein-3 and does not cross-react with other members of the Desmoglein-family. DSG3 is a calcium-binding transmembrane glycoprotein component of desmosomes in vertebrate epithelial cells. Research has shown that DSG3 has a very high sensitivity (80%) and specificity (100%) in recognizing squamous cell carcinoma (SqCC). Therefore, DSG3 is considered a very important marker for lung SqCC and can be a useful ancillary marker to separate SqCC from other subtypes of lung cancer. Gastrin, is a hormone that normally formed by mucosal cells in the gastric antrum and by the D cells of the pancreatic islets. Its primary function is to stimulate secretion of HCl by the gastric mucosa. HCl, in turn, inhibits gastrin formation. It also responsible for stimulating smooth muscle contraction and increasing blood circulation and water secretion in the stomach and intestine. Gastrin is regulated by epidermal growth factor in both mice and humans. Gastrin is excreted in excess by pancreatic tumors in the Zollinger-Ellison syndrome. Gastrin-Releasing Peptide (GRP) stimulates the release of gastrin as well as other gastrointestinal hormones and also acts as an autocrine growth factor for certain cell types. Recognizes a sialoglycoprotein of 39 kDa, identified as glycophorin A (GPA). It is present on red blood cells (RBC) and erythroid precursor cells. It has been shown that glycophorin acts as the receptor for Sandei virus and parvovirus. Glycophorins A (GPA) and B (GPB), which are single, trans-membrane sialoglycoproteins. GPA is the carrier of blood group M and N specificities, while GPB accounts for S and U specificities. GPA and GPB provide the cells with a large mucin like surface and it has been suggested this provides a barrier to cell fusion, so minimizing aggregation between red blood cells in the circulation. Reacts with human integrin beta3 (GPIIIa, vitronectin receptor beta chain). It associates with the IIb-chain (CD41) to form the GpIIb/GpIIIa complex (CD41/CD61). The CD41/CD61 complex appears early in megakaryocyte maturation. The activated CD41/CD61 complex is a receptor for von Willebrand factor, soluble fibrinogen, fibronectin, vitronectin and thrombospondin. It plays a central role in platelet activation and aggregation. The CD51/CD61 is implicated in tumor metastasis and adenoviral infection. Recognizes a protein of 30 kDa which is identified as Prohibitin, an evolutionarily conserved protein with homologues found in yeast to man. It is located in the inner membrane of mitochondria. Although prohibitin mRNA and protein expression occurs throughout the cell cycle, maximum levels are detected during the G1/S phase transition and minimum levels are seen in S phase and the G2/mitosis boundary. Prohibitin is located exclusively in the mitochondria with the highest concentration on the inner membrane. Prohibitin is an ideal mitochondrial marker. NKX3. 1 is a prostate specific gene encoding a transcription factor that plays an important role in normal prostate development and carcinogenesis. It is a prostatic tumor suppressor gene located on chromosome 8p21. 2, which frequently undergoes a loss of heterozygosity. NKX3. 1 expression is highly restricted in prostate epithelial cells and therefore can be used as a diagnostic biomarker for prostate cancer and other metastatic lesions of prostatic origin. Furthermore, NKX3. 1 shows better sensitivity than Prostate Specific Antigen (PSA) for identifying metastatic prostatic adenocarcinoma. This antibody recognizes a protein of 42 kDa, which is identified as Alpha-methylacyl-CoA Racemase (AMACR), also known as p504S. It is an enzyme that is involved in bile acid biosynthesis and beta-oxidation of branched-chain fatty acids. AMACR is essential in lipid metabolism. It is expressed in cells of premalignant high-grade prostatic intraepithelial neoplasia (HGPIN) and prostate adenocarcinoma. The majority of the carcinoma cells show a distinct granular cytoplasmic staining reaction. AMACR is present at low or undetectable levels in glandular epithelial cells of normal prostate and benign prostatic hyperplasia. The translocation of this MALT1 gene, which maps to human chromosome 18q21, and the apoptosis-inhibiting API2 gene results in an increased development of MALT lymphomas and apoptosis inhibition. The immunogen for this Mab is derived from the extracellular domain of CD3 epsilon chain. CD3 consists of five different polypeptide chains (designated as gamma, delta, epsilon, zeta, and eta) with MW ranging from 16-28 kDa. The CD3 complex is closely associated at the lymphocyte cell surface with the T cell antigen receptor (TCR). Reportedly, CD3 complex is involved in signal transduction to the T cell interior following antigen recognition. The CD3 antigen is first detectable in early thymocytes and probably represents one of the earliest signs of commitment to the T cell lineage. This gene encodes a receptor that binds the Fc region of monomeric immunoglobulin G.
