Reversibly inhibit caspase-3 and caspase-7 with Ac-DEVD-CHO (Garcia-Calvo et al. J Biol Chem 1998 Talanian et al. J Biol Chem 1997). Ac-DEVD-CHO is a caspase inhibitor with the PARP cleavage site DEVD which blocks PARP cleavage activity and inhibits caspase-3 and caspase-7 in a reversible manner (Garcia-Calvo et al. J Biol Chem 1998 Talanian et al. J Biol Chem 1997). This inhibition is via the aldehyde group’s interaction with the active site cysteine of these caspases (Thornberry et al. Perspect Drug Discov Des 1995). Caspase inhibitors are used to investigate caspase activation and apoptosis. Ac-DEVD-CHO is also used in cancer research to block camptothecin-induced apoptosis in bone tumor cells.
