Description

Parkin has been mapped to chromosome 6 and encodes a protein of 465 amino acids with similarity to ubiquitin at the amino terminus and a RING-finger motif at the carboxyl terminus. Parkinson’s disease is the second most common neurodegenerative disease following Alzheimer’s. The characteristic movement disorder results from dysfunction and neuron loss in the substantia nigra. Deletions in parkin, have been associated with autosomal-recessive juvenile parkinsonism.

Heat-mediated antigen retrieval is recommended prior to staining, using a 10mM citrate buffer, pH 6.0, for 10 minutes followed by cooling at room temperature for 20 min. Following antigen retrieval, incubate samples with primary antibody for 30 min at room temperature. A suggested positive control is Alzheimer's brain tissue.
