Rat Monoclonal Antibody

Description: The monoclonal antibody 5B10 recognizes mouse Caspase-1, a member of the cysteine-requiring aspartate protease family, which plays a critical role in cell suicide known as apoptosis. In particular, it converts pro-IL-1beta to mature IL-1beta; hence it is also called ICE (interleukin-1beta converting enzyme). Upon activation Caspase-1 exists as a tetramer of two p20 and two p10 subunits. There are several isoforms of Caspase-1. The 5B10 antibody recognizes both the pro-form of about 42kDa and the active p20 form. Applications Reported: This 5B10 antibody has been reported for use in immunoblotting (WB). Applications Tested: This 5B10 antibody has been tested by western blot analysis of lysates prepared from the J774 cell line. This can be used at 1-5 μg/mL. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest. Purity: Greater than 90%, as determined by SDS-PAGE. Aggregation: Less than 10%, as determined by HPLC. Filtration: 0.2 μm post-manufacturing filtered.

Caspases are a family of cysteine proteases that can be divided into the apoptotic and inflammatory caspase subfamilies. Unlike the apoptotic caspases, members of the inflammatory subfamily are generally not involved in cell death but are associated with the immune response to microbial pathogens. Members of this subfamily include caspase-1, -4, -5, and -12 and can activate proinflammatory cytokines such as IL-1-b and IL-18. Caspase-1 was initially identified as an IL-1-b-converting enzyme; later experiments revealed it to be a mammalian homolog of the C. elegans cell death gene ced-3 whose overexpression can induce apoptosis in fibroblasts.
