Rat Monoclonal Antibody

Description: The SER-4 monoclonal antibody reacts with mouse CD169, which is also known as Sialoadhesin and Siglec-1. CD169 is a 185-kDa member of the Siglec family of adhesion and endocytic receptors that recognize sialylated glycans. This receptor is expressed on a subset of macrophages that are found with high frequency in the spleen, liver, lymph node, and bone marrow. CD169 can also be detected on monocytes in the bone marrow and periphery. This receptor plays a role in cell-cell adhesion, as well as internalization of sialylated virus and bacteria. This endocytic pathway allows CD169+ macrophages to present lipid antigens to iNKT cells, thereby leading to their activation. Studies have also demonstrated that direct interaction of CD169 with regulatory T cells inhibits their proliferation. The SER-4 antibody has been reported to have antagonist activity. Applications Reported: This SER-4 antibody has been reported for use in flow cytometric analysis. Applications Tested: This SER-4 antibody has been tested by flow cytometric anaylsis of mouse bone marrow cells. This can be used at less than or equal to 0.25 μg per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

The siglecs are a family of membrane bound lectins (of the immunoglobulin superfamily) that bind sialic acid and mediate cell-cell interactions. Family members include sialoadhesin, CD22 and CD33. Sialoadhesin is a macrophage-specific cell adhesion molecule that is expressed on activated macrophages in chronic inflammation and in tumours.
